Long-chain Carnitine Acyltransferase and the Role of Acylcarnitine Derivatives in the Catalytic Increase of Fatty Acid Oxidation Induced by Carnitine.

چکیده
Carnitine-H3 or palmitate-U4 incubated with heart muscle preparations waa incorporated into a compound that had chromatographic behavior in several systems identical to that of palmitylcarnitine chemically synthesized from palmityl chloride and carnitine. Palmitylcarnitine biosynthesis from palmitic acid and carnitine was dependent upon ATP and CoA in addition to substrates and an enzyme preparation. In contrast, palmityl carnitine formation from palmityl CoA and carnitine did not require ATP or CoA. The following reaction, catalyzed by palmitylcarnitine transferase, was shown to be reversible: Palmitylcarnitine + Cod S palmityl CoA + carnitine.

متن کاملIn support of the roles of malonyl-CoA and carnitine acyltransferase I in the regulation of hepatic fatty acid oxidation and ketogenesis.
The rate of fatty acid synthesis in hepatocytes from meal-fed rats was manipulated over a wide range using glucose, lactate, and pyruvate to drive the system maximally, and glucagon, 5-(tetradecyloxy)-2-furoic acid (RMI 14,514), or a combination of both agents to inhibit lipogenesis. Measurements were made of cellular malonyl-CoA levels, long chain acylcarnitine concentration, and [l-‘4C]oleate...
متن کاملProperties of partially purified carnitine acetyltransferase.
Subsequent reports from our laboratory showed that both carnitine (/3-hydroxy-y-trimethylammonium butyrate) and acetylcarnitinei increased oxygen uptake and oxidation of long chain fatty acids by heart homogenates incubated under conditions optimal for fatty acid oxidation (2, 3). Acetylcarnitine action could not be attributed simply to formation of carnitine via hydrolysis because acetylcarnit...
متن کاملThe effects of ginsenoside Rb1 on fatty acid β-oxidation, mediated by AMPK, in the failing heart
Objective(s): This study intended to investigate the effects of Ginsenoside-Rbl (Gs-Rbl) on fatty acid β-oxidation (FAO) in rat failing heart and to identify potential mechanisms of Gs-Rbl improving heart failure (HF) by FAO pathway dependent on AMP-activated protein kinase (AMPK). Materials and Methods: Rats with chronic HF, induced by adriamycin (Adr), were randomly grouped into 7 groups. Gs-...

1. The induction of peroxisomal beta-oxidation activities by bezafibrate in cultured rat hepatocytes and in the rat in vivo was prevented by inhibitors of carnitine acyltransferase, e.g. 2-bromopalmitate, 2-[5-(4-chlorophenyl)pentyl]oxirane-2-carboxylate or 2-tetradecylglycidic acid. 2. The prevention of peroxisomal proliferation by carnitine palmitoyltransferase inhibitors could not be account...
